Product Title:
Dispase II, Neutral Protease, 20 x

Dispase® is a neutral protease which cleaves fibronectin, collagen IV, and to a lesser extent collagen I, and can be used to separate epithelial tissues from underlying mesenchyma.

Product Use:

CnT-DNP-10 can be used for the preparation of cells from a wide variety of different tissues and organs. It has been shown to be rapid and effective, yet gentle, when separating epithelial layers from their underlying substratum (i.e. the epidermis from the dermis in skin) during isolation procedures. It can also be used to subculture cells and prevent unwanted clumping of cells cultured in suspension.

Working concentration: Dilute to 1 x solution using the recommended culture medium.

On receipt, CnT-DNP-10 should be aliquoted and re-frozen (see below).

For isolation protocols, please see our resources section.

After 20 x dilution, CnT-DNP-10 yields 20 x 10 mL aliquots, or up to 20 isolation procedures.

Each lot of CnT-DNP-10 is tested during primary cell isolation, and the final concentration adjusted as necessary to deliver optimal performance.

Pack Size:

10 mL solution, 20 x

Comments:

Inhibitors: EDTA; EGTA; Hg2+; heavy metals; not inhibited by serum

Activators: Ca2+; Mg2+; Mn2+; Fe2+; Fe3+; Al3+

pH optimum: 6 to 8.5

Activity / Concentration:
The final 1 x solution has an activity of approximately 2.4 U / mL
Storage / Shelf Life:

Store at -20°C. See label for expiry date.

After initial thawing, CnT-DNP-10 should be aliquoted into desired quantities (e.g. 0.5 mL), and the aliquots refrozen and stored at -20°C for later use. Avoid repeated freeze and thaw cycles.

Thaw aliquots at at 4°C, and store at this temperature until use.   Use within 2 weeks of thawing.  Dilute to 1 x using recommended culture medium just prior to use.

Quality Control:

Tested for sterility and successful isolation of primary epithelial cells.

Shipping Conditions:

CnT-DNP-10 is shipped frozen.
