3D Keratinocyte Starter Kit including Human Primary Keratinocytes
3D keratinocyte starter Kit was developed for the establishment of 3 dimensional keratinocyte cultures on a porous membrane. Primary cells are cryo preserved at P2 and supplied with one 500 mL kit of CnT-57 medium (basal medium with frozen supplements), two 100 mL kits of CnT-02-3DP1 (basal medium with frozen supplements), stain solution and Millicell cell culture inserts.
The cells are of human origin and should therefore be handled as hazardous. Although it has been tested negative for HIV 1, Hepatitis B and Hepatitis C, treat this material as potentially infectious, and use appropriate biocontainment, protective equipment and other precautions to prevent accidental exposure.
Keratinocytes: Human, pooled (3 or more donors), neonatal foreskin
Kit is developed for human keratinocytes but may be used for other species as well.
3D-HPEK-25 is a kit consisting of:
- 1 x HPEKp.05.C: Human Epidermal Keratinocyte Progenitors > 5 x 105 viable cells per vial (1 mL)
- 1 x CnT-57: 500 mL kit of CnT-57 PCT Epidermal Keratinocyte Medium, Low BPE
- 2 x CnT-02-3DP1: two 100 mL kits of CnT-02-3DP1 Epidermal Keratinocyte 3D Prime Medium, Defined
- 25 x Millipore Millicell-PCF inserts: standing cell culture inserts, polycarbonate membrane, 12 mm diameter, 0.4 µm pore size, sterile and individually blistered
- 1 x CnT-ST: stain solution with 3 mL stain solution I and 3 mL stain solution II

All media are designed for use in a 5% CO2 atmosphere.

Keratinocytes guaranteed to provide 20 population doublings when grown in a PCT medium.
Immediately upon arrival transfer the cryo vial to the liquid nitrogen container, until ready to use.
Medium storage: basal medium (BM) at 4°C in the dark, supplements (S) at -20°C. For expiry date refer to medium label.
Supplemented medium: 4°C in the dark, use within 8 weeks of supplementation. Do not freeze.
Important: The quality of cell culture media is rapidly degraded by light. Store in the dark, and minimize all light exposure during handling.
Millicell-PCF inserts: room temperature. One year from date of purchase.
CnT-ST: room temperature. One year from date of purchase.
Recommended seeding density after thawing: 2 to 4 x 103 cells / cm2.

Tested negative for Hepatitis B, Hepatitis C and HIV-1. Free of bacteria, fungi and mycoplasma contamination.
Cells and supplements are shipped on dry ice.
Basal medium, Millicell-PCF inserts and stain solution are shipped at ambient temperature.

If desired antibiotics / antimycotics can be added.
