Epidermal Keratinocyte 3D Medium
CnT-02-3D5 is a liquid culture medium kit including both basal medium and separate supplements. CnT-02-3D5 is a fully defined, high calcium (1.2 mM) formulation. It does not contain antibiotics / antimycotics. The basal medium contains amino acids, minerals, vitamins and organic compounds.
The formulation of CnT-02-3D5 has been improved, leading to our new CnT-02-3DP5 3D Epidermal Keratinocyte Prime Medium. We strongly recommend to use the new formulation of CnT-02-3DP5 (same price).
CnT-02-3D5 is no longer listed in our product catalog. However, if you still need it for your work and do not wish to change to the newer version, CELLnTEC can still produce it for you.
CnT-02-3D5 is intended for use with CELLnTEC Primary Keratinocytes or your own keratinocytes isolated and grown in CELLnTEC PCT media (CnT-07 or CnT-57). Successful 3D growth requires a cell culture with a viable subset of undifferentiated progenitor cells. Keratinocytes isolated in non-CELLnTEC media may not contain enough of these cells to maintain the growth of a full thickness 3D model. Use of keratinocytes isolated and grown in other vendor’s medium may prove unsuccessful.
CnT-02-3D5 is a 500 mL kit consisting of:
- CnT-02-3D5.BM: 500 mL basal medium (liquid)
- CnT-02-3D5.S: containing three supplements (CnT-02-3D5.A, CnT-02-3D5.B and CnT-02-3D5.C; delivered frozen; to be added to 500 mL basal medium prior to use)
Developed for 3D (air / liquid interface) culture of epidermal keratinocytes on a porous membrane. Recommended membrane: Millipore Millicell Cell Culture Insert, PCF inserts, 0.4 μm. Cells should be established on the membrane in CnT-57 or CnT-07 medium, and then switched to CnT-02-3D5 medium after reaching confluency. Developed for 3D differentiation of human keratinocytes. May be used for other species as well (not yet tested).

CnT-02-3D5.BM basal medium: Store at 4°C in the dark. For expiry date, please see label on bottle. Please do not freeze.

CnT-02-3D5.S supplements: Store at -20°C. For expiry date, please see label on tube and bag.

The complete medium (basal medium supplemented with the corresponding supplements) must be stored at 4°C in the dark. Storage in this manner will provide a minimum shelf life of 8 weeks. Please do not freeze.
Important: The quality of cell culture media is rapidly degraded by light. Store in the dark, and minimize all light exposure during handling.
Tested by 3D cell cultivation in supplemented medium using CELLnTEC Kit and Protocols. Osmolality, pH and concentration of various ions are tested. Free of bacteria, fungi and mycoplasma contamination.
